The science of fasting

Autophagy: how many hours of fasting you need to trigger it

Autophagy is the cellular recycling that made fasting famous. When does it really kick in? No myths, no hype.

Part of the complete guide What happens in your body when you fast: benefits and science

What the science saysHow Many Hours of Fasting Triggers Autophagy?

There's no exact hour at which autophagy "switches on": it's a gradual process, not a switch. In mice it rises clearly after a short fast; in humans it has barely been measured directly, and the few available data points show changes in autophagy markers with daily 18-hour fasts. Most of the evidence is still animal research.

  • Eating only between 8 am and 2 pm (an 18-hour fast) for 4 days increased expression of the autophagy gene LC3A in 11 adults (Jamshed 2019). [1]
  • In mice, a short fast causes a marked increase in neuronal autophagy, along with lower mTOR activity (Alirezaei 2010). [2]
  • Fasting and calorie restriction are the most potent non-genetic inducers of autophagy; the evidence is mostly animal (Bagherniya 2018). [3]

What autophagy actually is

Autophagy (Greek for "self-eating") is the process your cells use to recycle old or damaged parts and put them back to work. It's cellular housekeeping. It earned Yoshinori Ohsumi the Nobel Prize in Medicine in 2016, and it's one of the reasons fasting gets linked to longevity.

How many hours until autophagy starts?

Time for some honesty: there's no exact switch. Most of the evidence (much of it in animals) suggests autophagy starts ramping up noticeably somewhere around the 16-18 hour mark and keeps climbing toward 24-48 hours. It's not like it turns on at 16:00 and stays off at 15:59.

It depends on your metabolism, your last meal, and how active you are. Think of a gradient, not a button.

The phases leading up to autophagy

HoursWhat's happening
0-4hDigestion. Insulin is high.
4-12hGlycogen runs down.
12-16hLight ketosis: you start burning fat.
16-24hAutophagy ramping up. Cellular cleanup.
24-48hAutophagy peaks, growth hormone rises.

How to boost autophagy

  • Extend your fast now and then (18-24h) if your health allows it.
  • Keep calories out of the fast: protein or sugar shuts it down. Black coffee is fine.
  • Exercise and good sleep stimulate it too.

Watch out for the myth

The internet oversells this. Autophagy doesn't "cure" everything, and you don't need a 3-day fast every week. Regular intermittent fasting already delivers benefits. More isn't always better, and very long fasts call for good judgment.

Frequently asked questions

How many hours of fasting does it take to trigger autophagy?
Autophagy starts ramping up noticeably around the 16-18 hour mark and keeps climbing toward 24-48 hours. There's no exact switch: it's a gradient that varies from person to person.
Does coffee break autophagy?
Black coffee doesn't break it, and it might even help slightly. What shuts it down is adding calories, especially protein or carbs.
Do I need very long fasts to get autophagy?
No. A regular 16/8 fast already stimulates it. Longer fasts push it further, but they aren't essential and they call for good judgment and solid health.
